Preparation and characterization of total flavones of hippophae rhamnoides (TFH) solid lipid nanoparticles by heating-ultrasonic

Abstract Purpose: To prepare TFH SLN (total flavones of hippophae rhamnoides solid lipid nanoparticles) and to investigate its characteristics. Methods: TFH SLN was prepared with heating-ultrasonic dispersion method and freeze drying technique was employed to get the dry powder. Results: TFH SLN dispersion is stable at pH 6.0–8.0, while its mean particle size of TFH SLN is about 200 nm. Fifteen% mannitol as the cryoprotective agents was able to protect nanoparticles and no significant changes in particle size of TFH SLN were observed after lyophilization. TFH SLN's relative bioavailability is about 175.2%. Conclusions: Crystal form of the drug and lipid matrix changed markably after being prepared into SLN, furthermore, TFH SLN has a longer releasing time and a good stability. TFH's relative bioavailability was improved a lot after being made into SLN.
